Scientists at the NYU Langone Medical Center and the Laura and Isaac Perlmutter Cancer Center are researching a form of cancer treatment called necroptosis (a form of orchestrated cell death) actually can increase the rate of production of CXCL, a small protein that can spur on the growth of PDAC tumor cells.
The CXCL1 protein can stimulate the creation of immunosuppressive cells, tumor-causing macrophages, which can numb the body’s ability recognize and in turn, kill the malignant cancer cells…
“Our findings are the first to show that cancer cell death via necroptosis can actually promote tumor growth, as this process results in suppression of the body’s immune response against the cancer,” says George Miller, MD, who was the leader of the investigation and also is the associate professor at th e Departments of Surgery and Cell Biology. “What is equally significant is that these findings might also be relevant to other tumor types.”
Something else they found was that the CXCL1 that was produced by the necroptosis, did not do enough when it attacked tumor’s environment and instead, when killed, the dying tumors released yet another protein, SAP130 which binds with Mincle, a receptor that is located on the cell membranes of the tumor cell’s enviroment… The binding then can cause even more tumors… The stimulation of the Mincle has been found to trigger faster growth of the tumors, leading to more deaths.
“This study exemplifies the importance of examining cancer within the actual context in which it grows,” said Gregor Werba, MD. “In our initial studies, inhibiting necroptosis in PDAC cells increased their ability to grow in tissue culture. However, when we began to study the same process in mice, we were surprised to see just the opposite effect, and this was mainly due to the immune response of the cells surrounding the tumor.”
Dr. Miller and his staff are helping Dierdre Cohen, MD, assistant professor at the Department of Medicine and also a prestigious member of the Perlmutter Cancer Center, in the investigation of possiblity this great cancer treatment and it’s thought that if used with Immunotherapy it could quite a deadly combination for cancer…
